Dosage form : Capsules , Sustained release : 10 mg , 20 mg and 40 mg.
Indications:
Short
treatment of active duodenal ulcer : 20mg daily for 4 to 8 weeks
For the relief of acid-related dyspepsia omeprazole is given in usual doses of 10 or 20 mg daily by mouth for 2 to 4 weeks.
treatment of gastro-oesophageal reflux disease is 20 mg by mouth once daily for 4 weeks
In the management of peptic ulcer disease a single daily dose of 20 mg by mouth, or 40 mg in severe cases, is given
Mechanism of action :
Omeprazole is a proton pump inhibitor. It inhibits secretion of gastric acid by irreversibly blocking the enzyme system of hydrogen/potassium adenosine triphosphatase (H+/K+ ATPase), the 'proton pump' of the gastric parietal cell
Interaction:
increase plasma levels of diazepam , phenytoin , and warfarin by inhibiting oxidative metabolism .
Omeprazole and other proton pump inhibitors can reduce the absorption of drugs such as ketoconazole, and possibly itraconazole, whose absorption is dependent on an acid gastric pH
Adverse effect:
back pain , cough , rash and dizziness
patient consultation :
Take 30 minutes prior to eating .
Antacid may be taken at the same time .
